Promotions, no.
For checking moves I don't know, but if you don't do anything special when sorting them, I suspect not. Worth testing though.

It depends on how the rest of your move ordering is. Everything that would be tried before the killers could at best be a duplicat that erases a killer that could have been useful. For history it is not as detrimental to award a count to a move that does not deserve it.
